INTRODUCTION
Phosphoinositide dependent protein kinase-1 (PDK1) is a "master"
protein kinase in insulin and growth factor signalling. Stimulation of cells
with insulin and growth factors leads to the generation of
the phospholipid second messengers phosphatidylinositol (3,4,5) trisphosphate
(PtdIns(3,4,5)P3) and PtdIns(3,4)P2 through activation of PI3 kinase. These
second messengers trigger the activation of a complex protein kinase network,
which is responsible for basic cellular processes such as cell growth and cell
division. The Ser/Thr protein kinase 3-phosphoinositide dependent protein kinase
1 (PDK1)
is responsible for the activation of at least 24 different protein kinases in
this network, such as protein kinase B (PKB), p70 S6 kinase (S6K), and serum and
glucocorticoid responsive kinase (SGK). PDK1 phosphorylates its substrate
kinases in their T-loop (also called activation segment), which leads to the
activation of the respective kinase. Because of it's early and central role in
this signalling network, PDK1 has been termed a "master kinase". Sequence
analysis predicted that the 556-amino acid PDPK1 protein contains a catalytic
domain with 11 classic kinase subdomains and a C-terminal pleckstrin homology
